WT-Ovation™ Pico System
The WT-Ovation™ Pico RNA Amplification System provides a sensitive, rapid, and simple method for preparing in less than five hours six to 10 micrograms of amplified cDNA from a range of 500 picograms to 50 nanograms total RNA.. This product is specifically designed with very low RNA input requirements and enables detection of transcript in a wide range of abundance. Amplification is initiated both at the 3' end and randomly throughout the whole transcriptome to enable amplification of non-poly(A) transcripts and compromised RNA samples, and to provide flexibility in qPCR assay design.
The amplified cDNA can be fragmented and labeled in less than two hours for GeneChip® 3’ expression array analysis using NuGEN’s FL-Ovation™ cDNA Biotin Module V2.
For analysis using Affymetrix GeneChip® Exon and Gene ST arrays, the amplified cDNA can be used with the WT-Ovation Exon Module, then fragmented and labeled using the FL-Ovation™ cDNA Biotin Module V2.
For analysis on Agilent arrays, the amplified cDNA can be fragmented and labeled using NuGEN’s FL-Ovation™ cDNA Fluorescent Module.
The WT-Ovation™ Pico RNA Amplification System provides optimized reagent mixes and a protocol to process 12 total RNA samples (cat.# 3300-12).

Colon total RNA were amplified in triplicate with a wide range of input using the WT-Ovation™ Pico RNA Amplification System. Five qPCR assays were designed and performed for three transcripts and Ct values were normalized to the PGK transcript. qPCR results are shown with Delta Ct plotted against the RNA input and demonstrate that the Ct values from each set of transcripts remain consistent across a wide range (100 pg to 50 ng) of total RNA input for all interrogated transcripts, even at RNA input levels far below the recommended range.
